چکیده

The effects of monovalent (Na, K) and divalent (Mg, Ca, Mn) ions on the interaction between DNA and histone are studied using the molecular combing technique. λ-DNA molecules and DNA-histone complexes incubated with metal cations (Na, K, Mg, Ca, Mn) are stretched on hydrophobic surfaces, and directly observed by fluorescence microscopy. The results indicate that when these cations are added into the DNA solution, the fluorescence intensities of the stained DNA are reduced differently. The monovalent cations (Na, K) inhibit binding of histone to DNA. The divalent cations (Mg, Ca, Mn) enhance significantly the binding of histone to DNA and the binding of the DNA-histone complex to the hydrophobic surface. Mn also induces condensation and aggregation of the DNA-histone complex.

Molecular Effects of Atmospheric Pressure Plasma Jet on the Double-Stranded DNA

Introduction The aim of this study was toinvestigate the sterilization potential of atmospheric pressure plasma jet (APPJ) and interactions of this technology with double-stranded DNA using the polymerase chain reaction (PCR) and single-strand conformation polymorphism (SSCP) techniques. Materials and Methods The plasma jet was produced through a high voltage sinusoidal power supplyusing a mixt...

The effect of aspirin on the interaction of histone 05 and 05-DNA

The linker histones (H1 or H5) which play a key role in the folding of chromatin, are general repressors of gene expression. Nuclei of the mature chicken erythrocytes (and in some mammalian cells) contain both of them. Although the interaction of H5 with DNA is stronger than that of H1, it does not prevent the transcription of some erythroid-specific genes. It has been shown that some modificat...

Ionic effect on combing of single DNA molecules and observation of their force-induced melting by fluorescence microscopy.

Molecular combing is a powerful and simple method for aligning DNA molecules onto a surface. Using this technique combined with fluorescence microscopy, we observed that the length of lambda-DNA molecules was extended to about 1.6 times their contour length (unextended length, 16.2 microm) by the combing method on hydrophobic polymethylmetacrylate coated surfaces. The effects of sodium and magn...
